NOW Flush-Free Niacin 250 mg is a mono-preparation based on inositol hexanicotinate, a chemical compound supplementing a diet in large amounts of niacin without causing the “flush” effect, which is troublesome for many people, giving symptoms of redness and itching of skin. Thanks to providing the organism with a large portion of niacin, it ensures the possibility of unwavering course of many biochemical processes, especially energy processes, supports antioxidant barrier, reduces the size of inflammations and considerably takes care of maintaining proper functionality of cardiovascular and nervous system, which considerably improves comfort of life. The product appears in the form of cellulosed capsules and is also suitable for vegans.
Niacin – also called vitamin B3 or PP, a common name for a group chemical compounds, demonstrating biologically similar effects, to which we classify: nicotinic acid, its amide form – nicotinamide and recently discovered nicotinamide riboside. Among the most characteristic features there are good solubility in water and alcohol and generally high resistance to high temperature, in comparison to other vitamins, reaction fluctuations or light. It was synthesized for the first time in 1867, however, as late as in the XX century its beneficial properties were noticed. In the human organism the most reactive form is nicotinic acid, which for higher safety circulates in the organism in the form of active amide, of which tissues more willingly take advantage in numerous transformations. When needed, nicotinic acid is restored to the form of amide in the process catalyzed by deamidase of intestinal microflora or liver. In biochemical reactions, niacin is used mainly as a substrate for the synthesis of nicotinamide adenine dinucleotide (NAD+) and its phosphorylated form nicotinamide adenine dinucleotide phosphate (NADP+), coenzymes having considerable influence on the multitude of processes in the organism. Direct influence on appropriate molecules, including nicotinic acid receptors, also has influence on the organism. The main food products rich in niacin are: yeasts, meat, especially white meat, milk, eggs, fish, whole seeds, red pepper, green vegetables and tea. When needed, the human organism demonstrates the ability of endogenous synthesis of niacin from tryptophan, where by using 60 mg of a given amino acid, it is able to produce 1 mg of a desired vitamin. The dose of niacin obtained from food, essential to maintain continuity of biochemical processes in the organism are the values between 15-20 mg.
Properties of niacin
Required for anabolic and catabolic processes
By constituting a precursor of nicotinamide adenine dinucleotide (NAD+), it ensures the organism the ability to perform many catabolic processes, including those of carbohydrates, proteins or fats. Being essential in the production of the phosphorylated form of nicotinamide adenine dinucleotide (NADP+), it is engaged in the occurrence of anabolic processes, mainly regarding lipids. By promoting the release of the growth hormones, it supports the development of many structures, mainly protein.
Increases the capacity of antioxidant barrier of the organism
Thanks to NADPH accompanying the activity of glutathione reductase (GSH), it supports regeneration of glutathione and its transformation to the reduced form, as a result of which the aggressiveness of hydrogen peroxide (H2O2) in the organism is lower. It noticeably limits lipid peroxidation and promotes the protection of autosomal antioxidants. The effects approximately exceed antioxidant ability of β-tocopherol.
Demonstrates antioxidant effect
By inhibiting the activity of poly (ADP-ribose) polymerase-1 (PARP-1), it slows down the production of numerous inflammatory substances, including: cytokines, chemokines or adhesive molecules. By decreasing excitability of macrophages, limiting the pace of transformations of lymphocytes, decreasing the activity of macrophages and the synthesis of pro-inflammatory cytokines, e.g. IL-1, IL-6, IL-8, IL-12, TNF-α and ICAM-1, it reduces inflammations in the organism.
Is engaged in energy metabolism of the organism
By increasing the access of substrates of the complex I, it supports maintaining the required energy metabolism in mitochondria. By interfering with the transformations of lipids, proteins and carbohydrates, it manages energy substrates.
Has neuroprotective properties
By having antioxidant and anti-inflammatory effect, it supports maintaining conditions, stabilizing the structures of the nervous system. By promoting maintaining the required stimulation of Δ-6-desaturase, decreasing the activity of caspase 3 and limiting the release of cytochrome c from mitochondria, it supports the protection of neurons from their premature destruction. In the period of the sudden deterioration of conditions in the central nervous system, it considerably increases the level of VEGF activating PI3K/Akt and it supports the creation of vessels in the brain in the period of the improvement of the conditions. Moreover, it influences the decrease of TNF-α. By maintaining integrity of structures of the nervous system and accelerating its renewal, it is engaged in maintaining proper functionality of a given system and good psychological state, resulting from unwavering work of the central nervous system.
Decreases migraine headaches
By increasing the synthesis of prostaglandin D2 (PGD2), it supports widening of intracranial vessels and inhibits narrowing of extracranial vessels, which leads to stopping severe headaches. Moreover, by supporting blood flow with oxygen to the brain and reducing the amount of lactate and pyruvate, it decrease the frequency of the appearance of headaches in people susceptible to this kind of pain.
Maintains good condition of the cardiovascular system
By increasing effectiveness of antioxidant barrier and by having antioxidant effect, it protects endothelial cells from damages. By decreasing the amount of free radicals and prolonging the survival of endothelial nitric oxide, it supports widening of vessels. By increasing the activity of endothelial pathway COX-2/PGI2, it has anticoagulant effect. By having considerable influence on fat transformations, it is conducive to optimization of lipid profile, which is reflected in lowering the level of triglycerides, total cholesterol, low density lipoproteins (LDL), including lipoproteins (a) as well as increasing the level of high density lipoproteins (HDL). It demonstrates additional care of the condition of vessels thanks to intensifying the excitability of adiponectin, a hormone responsible for the transformations of glucose and fatty acids, demonstrating certain anti-inflammatory and protective properties towards cells.
Takes care about proper state of kidneys
By decreasing the expression of NaPi/2b transporter in kidneys, it reduces the size of absorbed phosphates, thanks to which it reduces the burden of kidneys and limits the creation of vessel calcifications, which supports the work of this organ.
Is conducive to maintaining good conditions of pancreas
By reducing the amount of reactive oxygen species produced by macrophages, decreasing the expression of iNOS stimulated by the presence of cytokines and lowering the activity of PARP, it counteracts degradation of beta cells in the islets of Langerhans, which considerably influences maintaining the required functionality of the pancreas.
Promotes maintaining proper state of the stomach
By demonstrating antioxidant activity and increasing the amount of mucus covering the stomach in the period of increased susceptibility to toxins, e.g. alcohol, it protects the structures of the stomach, demonstrating gastroprotective effect.
Long-lasting deficiency of niacin in a diet may lead to the occurrence of skin changes, such as: dryness and redness in the places exposed to sunrays and injuries, blisters and ulcers or skin discolorations; it also results in the worse quality of mucosa of the digestive system and its general functionality and deepening of inflammations in the nervous system as well as disorders in psychological balance. An extreme consequence of niacin deficiency is pellagra, a disease, which combines all the above symptoms, for the first time described in 1735, which was caused in that time by very low supply of meat in a diet and large consumption of easily available corn, poor in tryptophan.
Supplemented forms of niacin
The two simplest supplemented forms of niacin are nicotinic acid and nicotinamide, which is an amide form of nicotinic acid. The majority of properties identified with niacin are demonstrated by nicotinic acid, characterized by fast absorption and immediate release and effect. However, it causes some symptoms in the form of redness and hot flashes, called the “flush” effect, which may be troublesome for people taking this form of the vitamin. There is a possibility to decrease the inconveniences by using a form with prolonged or long-lasting release, although these forms should be taken with caution, as they are considered harmful for the liver. Nicotinamide, in turn, does not lead to the creation of the above mentioned effect and presents a bit modified properties attributed to niacin, when it comes to strength, including lower effectiveness in reducing cholesterol and triglycerides.
A perfect compromise between the strength of properties and toning the “flush” effect seems to be inositol hexanicotinate (IHN), which is the combination of inositol creating a scaffolding of the compound with six niacin molecules, combined with carboxyl bonds. This form is characterized by high level of bioavailability and lack of the “flush” effect after application. Admittedly, it does not seem to be so effective in optimizing lipid profile as nicotinic acid, however, it works well in facilitating the functioning of the circulatory system, which is reflected in the improvement of vessel expandability and blood stickiness as well as support of oxygen transport. Inositol molecule incorporated in this chemical compound ensures additional beneficial effects.
Inositol – also known as cyclohexane-1,2,3,4,5,6-hexol, a chemical compound from the group of polyols, a molecule structurally similar to glucose. Due to important properties for the human organism, it is called vitamin B8, although, from the nutritional point of view it is considered a pseudo-vitamin. It has nine isomers, among which the most popular is active myo-inositol. The effects caused by the presence of inositol are the result of its direct and indirect activity thanks to chosen hormones, i.a. insulin, gonadoliberin or thyrotrophic hormones (TSH).
NOW Flush-Free Niacin 250 mg is a product, providing large amounts of niacin, in a safe form of inositol hexanicotinate, which does not cause the “flush” effect. By providing a proper portion of niacin, it improves the work of energy metabolism in the organism, takes care of maintaining the required functionality of the nervous and circulatory system and strengthens antioxidant barrier, decreasing excessively spread inflammations. An additive of inositol, introduced with a cell constituting a carrier of niacin, constitutes the activity of niacin in the above mentioned aspects. Encapsulation of the product based on cellulose minimizes unbeneficial digestive discomfort caused by gelatin encasements and provides a possibility to take the vitamin in this form by vegans.
A daily portion contained in the preparation is relatively high, therefore I don’t recommend taking a higher portion that the one recommended by the producer. In order to promote cognitive functions, a daily dose (2 capsules) may be taken ca. one hour before the planned mental effort.
