BRAWN Arimestane-30 is a mono-preparation, demonstrating anti-estrogen and anti-cortisol effect, based on androsta-3,5-diene-7,17-dione, a compound also known as 3-deoxy-7-oxo-DHEA, or commonly arimistane, a substance structurally similar to 5-dehydroepiandrosterone.

5-dehydroepiandrosterone is an androgen called DHEA, or more precisely 5-DHEA. From the chemical point of view, it is a compound based on cyclopentanoperhydrophenanthrene, which classifies it to the group of steroid substances, also known as steroids. In the human organism, it plays the role of androgen, i.e. a steroid sex hormone with masculinizing effect. Its autosomal synthesis takes place primarily in the fibroreticular layer of adrenal cortex. Less prominent amounts are produced in testicles and ovaries. A starting compound that enables its production, similarly to other types of steroids, is cholesterol. Both autosomal production as well as the effects and scope of activity of this compound are quite spectacular. Maintaining proper DHEA level ensures appropriateness of the functioning of the cardiovascular, nervous, osteoarticular, reproductive and immune systems, as well as optimal activity of energy metabolism, mainly by the influence on managing energy substrates and it also ensures proper psychological state. The above mentioned roles in the organism are played directly or indirectly by means of metabolites of its transformations, i.e. testosterone, androstenedione and androstenediol. Directly and as a result of the activity of androstenediol, it decreases the activity of 11β-hydroxysteroid dehydrogenase type1 (11bHSD1), thanks to which it allows to decrease unbeneficial effects of previously increased cortisol level. By increasing the activity of androgen receptors (AR), located in muscles, it promotes their growth. By intensifying AR and receptors activated by peroxisome proliferator-activated receptors (PPAR), it supports proper management of energy substrates. As an androgen which does not undergo transformations to dihydrotestosterone (DHT), in the presence of 5α-reductase and as an androgen which does not strongly and directly influences AR placed in prostate, it does not contribute to its intensive hypertrophy. By being unsusceptible to the activity or aromatase and by activating estrogen receptors on its own only to a minimal degree, it does not lead to the negative consequences of excessive activity of female hormones.

Arimistane, being a derivative of DHEA, demonstrates many effects similar to its structural cousin. However, it demonstrates the most particular effects by quite a different way of controlling the process of autosomal synthesis of estrogens.

Estrogens are a few compounds from the group of steroid hormones, to which we classify: estradiol, estrone and estriol. They are directly synthesized from testosterone and androstenedione with the utilization of aromatase. Their largest amounts are produced in the ovaries, while estrone in the cells of adipose tissue. Due to their primary place of synthesis and playing a special role in the organisms of women, i.e. creating female features and participating in regulating menstrual cycle, estrogens are associated with female sex. Regardless of this, they fulfill many other functions in the human organism, which are also important for men, as they take part i.a. in the process of bone mineralization, protein assimilation or normalization of lipid profile. By stimulating 6-phosphate dehydrogenase and increasing the secretion of somatotropin (GH) and insulin-like growth factor 1 (IGF-1), it considerably supports the growth and regeneration of muscle tissue. Positive effects of the activity of estrogens are noticed, when their optimal concentrations are ensured, as the excessive increase of their amounts, especially in men, may lead to the occurrence of a range of unbeneficial effects, including excessive storage of fat, increased water retention, sleep problems, increased blood pressure and also breast hypertrophy - a disorders in men, called gynecomastia. 

Androsta-3,5-diene-7,17-dione by binding with aromatase and not undergoing its catalytic activity, strongly blocks the activity of aromatase. At the same time, it leads to inhibiting endogenous production of estrogens from testosterone and androstenedione circulating in the organism.

Anti-estrogen properties of Arimistane are even more important in the periods of abnormal concentration of free testosterone in the organism. Androsta-3,5-diene-7,17-dione, by protecting free testosterone from aromatization and at the same time supporting the secretion of the luteinizing hormone (LH) engaged in endogenous synthesis of natural testosterone, is conducive to safe maintenance of the high concentration of testosterone in the organism. At the same time, it promotes undisturbed activity of testosterone in the scope of intensifying muscle anabolism, at the same time minimizing the occurrence of unbeneficial side-effect of female hormones.

3-deoxy-7-oxo-DHEA is also an important anti-cortisol compound, thanks to which it demonstrates additional ability to increase safety of intensive development of muscle mass.

Cortisol, also, called hydrocortisone, is a dominant steroid in the group of glucocorticosteroids, playing the function of a hormone in the human organism. Endogenously, it is produced from 11-β-deoxycortisol in zona fasciculata of adrenal cortex. 11β-hydroxysteroid dehydrogenases type 1 and 2 are responsible for maintaining an appropriate level of their activity, while the former promotes the process of cortisol activation and the latter inactivates it by transforming it to cortisone. The activity of cortisol is noticed most intensively in stressful conditions, e.g. in physical and/or mental exhaustion or starvation diet. By intensifying the processes of glycogenolysis and gluconeogenesis and antagonizing the activity of insulin, it increases the amount of energy substrates in the bloodstream. By inhibiting the storage of energy substrates in the form of adipose tissue and slowing down their utilization by muscles, they ensure their better availability for other areas of the organism. It intensifies the transformation of fatty acids in ketone bodies. It extracts amino acids from protein structures, especially muscle tissue. It increases resorption of sodium, intensifying at the same time the excretion of potassium. It slows down building calcium in bone structure. It decreases the lumen of blood vessels. Although a certain level of cortisol is required to maintain proper work of the organism, its long-lastingly high concentration may develop some unbeneficial, pathological states, giving symptoms of, i.a.: general fatigue, drop of muscle mass, increase of insulin-resistance, disorders in the distribution of fat, stored especially in the area of the belly, chest, face and neck, excessive water retention, increase of blood pressure, weaker bones or deterioration of the work of the immune system.

Androsta-3,5-diene-7,17-dione by decreasing the level of cortisol decreases the feeling of stress and allows to create brighter thoughts and reactions which are adequate to a given situation.
