Seeking Health Vitamin D3+K2 is a dietary supplement ensuring the complex of vitamins demonstrating synergistic effect in ensuring proper bone mineralization – vitamin D3 at the dose of 5000 IU (in the form of cholecalciferol) and vitamin K2 MK-7 (as menachinone-7). Cholecalciferol has been obtained from ship wool, which makes the product appropriate for vegetarians.

Vitamin D3 (cholecalciferol) is a vitamin of steroid structure. It is produced from provitamins D3 (7-dehydrocholesterol) present in epidermis and dermis under the influence of UV-B radiation. It is transformed in the liver and kidneys to biologically active forms – calcidiol (25(OH)D3) and calcitriol (1,25(OH)D3).

Calcitriol has influence on target cells (i.a. epithelial cells of intestines, bones, heart, brain, beta cells of the pancreas) by means of nuclear receptors VDR, controlling the expression of more than 200 genes. Moreover, calcitriol demonstrates affinity to membrane receptors, activating in this way kinases, proteases and inducing the synthesis of prostaglandins.

Vitamin D3 is responsible for proper calcium-phosphorus balance, conditioning optimal mineralization of bones and at the same time preventing the development of osteoporosis. It regulates the immune response by the stimulation of decreasing inflammatory reactions. It is also essential for the proper functioning of the circulatory system. It increases the level of glutathione, a powerful antioxidant in the organism, protecting from the toxic activity of free radicals and facilitating liver detoxification.

Vitamin K belongs to the group of vitamins soluble in fats, quinone derivatives. In the group of vitamins K we may distinguish vitamin K1 (phylloquinone), K2 (menaquinone) and synthetic K3 (menadione). These forms are different in bioavailability, activity and partially in the influence on the organism. In general, vitamin K is the substance that conditions optimal blood coagulation, influencing the synthesis of prothrombin (thrombin precursor, factor II of coagulation) and factors VII, IX, X. It is also essential for the synthesis and modification of proteins taking part in building bones and also it demonstrates antibacterial effect.

Vitamin K2 (menaquinone), contrary to phylloquinone, it is redistributed by the liver and blood in the whole organism, thanks to which it is highly available for tissues, such as bones or blood vessel walls. Clinical research confirmed the effectiveness of vitamin K2 in reducing the frequency of bone fractures. A superior role of menaquinone is modification and activation of proteins taking part in building them – osteocalcin, transporting proteins, enabling storing calcium in bones instead of artery walls. Deficiency of vitamin K2, by its participation in regulation of calcium transport is a common denominator of osteoporosis and circulatory system diseases.

Osteocalcin is a biological marker of bone formation. It is synthesized by osteoblasts and osteocytes. Vitamin K2 influences the expression of osteoblastic markers of bones by the activation of SXR receptors (Steroid and Xenobiotic Receptor). Osteocalcin has the ability to bind calcium ions and maintain them in bones. In case of the lack of active osteocalcin (low level of vitamin K2), calcium is stored in the arteries, leading to blood vessel calcification and consequently the development of atherosclerosis and the increase of the risk of heart attack. Proper distribution of calcium is therefore incredibly important in preventing cardiovascular system diseases – ailments which are the main civilization diseases.
